چکیده
Staphylococcus aureus is a Gram-positive, round-shaped bacterium that member of the Firmicutes. It often positive for catalase and nitrate reduction. Pathogenic strains promote infections by producing virulence factors such as potent protein toxins, expression cell-surface binds inactivates antibodies. Sepsis-related to methicillin-resistant S. (MRSA) has significant morbidity high mortality rates (15-30%). The methicillin resistance coded with mecA gene, while sensitivity nuc they are chromosomal. Similarly, it coagulase gene (Coa). In this study, 16S rRNA identification Real-Time PCR was investigated in forty isolates, which were cultured at different times terms MIC SIR tests. isolates used study determined level their differences (MecA), susceptibility (Nuc), (Coa) intra-species examined. As result spp. yielded results gene-specific primers all isolates. analysis SYBRGreen-based performed primers, samples confirmed be Staphylococcus. Analysis family followed Coa, Nuc, MecA results, found that, Coa Nuc genes, 19 21 negative. 16 according sigmoidal curves single peak melting values, whereas 24 thought will benefit community contributing rapid effective diagnosis treatment caused negative Staphylococci.

Rapid detection of mecA-positive and mecA-negative coagulase-negative staphylococci by an anti-penicillin binding protein 2a slide latex agglutination test.
A rapid slide latex agglutination (LA) test, MRSA-Screen (Denka Seiken Co., Niigata, Japan), which detects PBP 2a, was tested for its ability to differentiate between mecA-positive and -negative coagulase-negative staphylococci. A total of 463 isolates from 13 species were included in the study. The mecA gene was detected by PCR, and the oxacillin MIC was determined by the agar dilution method ...
متن کاملDetermination of the chromosomal relationship between mecA and gyrA in methicillin-resistant coagulase-negative staphylococci.
mecA, the gene that mediates methicillin resistance, and its accompanying mec locus DNA, insert near the gyrA gene in Staphylococcus aureus. To investigate whether there is a similar relationship between mecA and gyrA in coagulase-negative staphylococci (CNS), mecA- and gyrA-specific DNA fragments were used to probe methicillin-resistant isolates of Staphylococcus epidermidis (MRSE) (n = 11) an...

متن کاملAntibiotic resistance and mecA characterization of coagulase-negative staphylococci isolated from three hotels in London, UK
Antibiotic resistance in bacteria isolated from non-healthcare environments, is a potential problem to public health. In our survey a total of 71 coagulase negative staphylococci (CNS) belonging to 11 different species were isolated from three large hotels in London, UK. The most prevalent species was Staphylococcus haemolyticus, with S. hominis, S. warneri, S. cohnii, and Staphylococcus epider...
متن کاملSimultaneous DNA-RNA Extraction from Coastal Sediments and Quantification of 16S rRNA Genes and Transcripts by Real-time PCR
Real Time Polymerase Chain Reaction also known as quantitative PCR (q-PCR) is a widely used tool in microbial ecology to quantify gene abundances of taxonomic and functional groups in environmental samples. Used in combination with a reverse transcriptase reaction (RT-q-PCR), it can also be employed to quantify gene transcripts. q-PCR makes use of highly sensitive fluorescent detection chemistr...
